RESULTNS1andC4BPbindingELISA1.ToaddresswhetherflavivirusesuseNS1tocontrolC4bactivationbyrecruitingC4BP,theyevaluatedifNS1boundhumanC4BPIncreasingionicstrengthofthebufferdidnotappreciablyaffecttheNS1–C4BPinteraction,suggestinganonionicinteractionbetweenC4BPandDENVorWNVNS1.CoimmunoprecipitationexperimentsconfirmedtheinteractionbetweenNS1andC4BPC4bbindingprotein(C4BP)hasalargespider-likestructureandiscomposedofsevenidentical70-kDaα-chainsandone45-kDaβ-chainthatarelinkedbydisulfidebondsattheirC-terminaldomainsofC4BPcontaineightandthreecomplementcontrolprotein(CCP)
